The drug undergoes rapid
metabolism in intestinal mucosa due to change in pH environment and hence decreased oral bioavalability. The aim of
present Investigation is to increase the gastric residence time by preparing gastro retentive mucoadhesive tablet here by
improving bioavalability of cefuroxime axetil. A simple UV spectrophotometric method has been employed for the
estimation of cefuroxime axetil at 280nm with a Beer’s range of 4-24'g/ml. Fourier transaform infrared spectroscopy
confermed the absence of any drug/polymer/excipient interaction. Tablets of various formulations were prepared using
different mucoadhesive polymers such as carbopol 974P, Hydroxypropylmethyl cellulose K4M, Sodium alginate, Chitosan
in optimized combination ratios by direct compression method. All the batches were subjected to various evaluation
parameters such as physicochemical properties, swelling characteristics, in vitro residence time, and in vitro drug release
profile. Optimized formulation containing carbopol 974P and chitosan in combination (F6) exhibited maximum in vitro
residence time of 10.05 hr and in vitro release up to 84.93% in 10 hours. This formulation was further subjected to in vitro
permeation, SEM studies, stability studies and in vivo residence time. Scanning Electron Microscopy (SEM) revealed
smooth surface characteristics with increasing pore diameter indicating the diffusion mechanism of release. Stability was
carried out as per ICH guidelines at 40±20C/75±5% RH and the values were within permissible limits. In vivo X-Ray
photographs taken during the experiment showed mucoadhesion to the gastric mucosa up to 10 hours.
<Email>drarshadomc@gmail.com</Email><articletype>Research</articletype><volume>6</volume><issue>5</issue><year>2016</year><keyword>Cefuroxime axetil,Gastroretentive,Mucoadhesive tablets,Polymers,X-ray study</keyword><AUTHORS>Syed Arshaduddin Ahmed, Syed Atiq-ur Rahman</AUTHORS><afflication>Department of Pharmacology, Osmania Medical Coleege, Hyderabad, Telangana, India.,Department of Pharmacology, Osmania Medical Coleege, Hyderabad, Telangana, India.</afflication></Article></Articles>
